This channel is about different aspects of Biology particularly Microbiology and Botany. All the videos are meticulously made to serve the deeper understanding of the subject. I have two principal purposes of making youtube videos - one for my own practice and the other to provide free knowledge to the knowledge hunters. But unfortunately the second purpose is being poorly served. Anyway, thank...
Ti-plasmid Vector | Agrobacterium mediated gene transfer | Leaf-disc method | vir genes

Ti Plasmid Vector - The Ti (tumor inducing) plasmid is naturally occurring plasmid found in the common soil borne gram negative bacterium called, Agrobacterium tumefaciens. This bacterium and a close relative of it, the Agrobacterium rhizogenes that posseses Ri (root inducing) plasmids are known to cause crown gall disease and hairy root formation respectively in dicots. The reason behind this ...
How to remember Restriction sites of some Restriction Endonucleases?
Просмотров 204 месяца назад
This video talks about how to remember some important restriction endonucleases and their recognition sites. The video, however, starts with Sticky end and Blunt end producers and how are they produced. This video may help students to keep this thing in mind for longer period of time. Dr. Sanjeev Pandey
How many EcoR1 sites may be found in a 10K DNA?
Просмотров 234 месяца назад
Thousands of restriction enzymes are known and the list is ever increasing. Most of these enzymes recognize sequences which are 4-8 basepairs long. Restriction endonucleases that recognize 4-bp long sequences are called tetracutters, those that recognize 6 bp are called hexacutters and those that recognize 8-bp long sequences are called octacutters. It is quite natural that the probability of f...
Restriction-Modification System | Restriction Endonuclease

Restriction Endonucleases (Restriction- Modification (RM) System) In nature there is a host-pathogen specificity and bacterial and phage relationship is no exception to it. Each phage has a specific bacterial host. Whether a phage will be able to infect and multiply within a host bacterium is governed by the restrictionmodification system (RM-system) the cell possesses. This became evident afte...
Adaptor Hypothesis | Significance of tRNA

Proteins are made of amino acids, that means, sequence of nucleotides in the mRNA are encoded into sequence of amino acid of a protein. In the last chapter we learnt that each amino acid of a protein is encoded by a triplet nucleotide sequence called genetic codon. Since amino acids are not complementary to the nucleotide sequences in the mRNA, there must be an adaptor that bridges this gap bet...
Which enzyme of our cells is Reverse Transcriptase | Telomerase
Просмотров 805 месяцев назад
Each and every eukaryotic cell contains an enzyme called telomerase for the synthesis of telomere. It is basically a reverse transcriptase with an RNA component having the complementary sequence of the repeat element of the telomere. Thus telomerase is an RNP (Ribonucleoprotein). The telomerase binds at the 3’ end of of the DNA telomere and extends it to telomeric repeat length each time using ...
DNA Polymerases require primers | RNA polymerases do not | Why?
Просмотров 1025 месяцев назад
The primer in cellular DNA replication is an RNA molecule was proved when an M13 phage DNA was allowed to replicate in E. coli cell lysate in presence of the antibiotic rifampicin. Replication was inhibited in presence of rifampicin which is an inhibitor of RNA polymerase. This gave the first indication that RNA polymerase (primase) is involved in the process of DNA replication. The RNA polymer...
Rolling Circle mode of DNA Replication
Просмотров 285 месяцев назад
Rolling circle model - This model was proposed by Gilbert and Dressler in 1968. Rolling circle mode of replication occurs in F-plasmid at the time of F × F- and Hfr × F- conjugation. It is also found to occur in λ-phage DNA at the time of packaging. Packaging requires a minimum of one and half cos site that is only possible if the DNA undergoes rolling circle mode of replication followed by con...
Theta Model of DNA replication
Просмотров 2105 месяцев назад
Theta (θ)model of DNA replication - As mentioned before, this model was proposed by John Cairns in 1963 during his studies with 3H-thymidine to understand the semiconservative nature of replication in E. coli. His experiments proved that the bacterial chromosomes replicate by this theta model of replication. However, theta model of replication also occurs in λ-phage DNA after its entry into the...
Cell division Replication Asynchrony
Просмотров 305 месяцев назад
The E. coli genome replicates in 40 minutes but the cells divide only in 20 minutes. The cells have to receive one chromosome. So, how can the have this when the replication process is yet not completed. The answer is the cell when got its genome from its parent cell, it had received a half replicated chromosome. So to complete the remaining portion 20 minutes is required, so it can deliver eac...
Cot value and Cot curve

Cot VALUE Developed by Roy Britten and his colleague during 1960s, Cot value is a measure of repetitiveness in a DNA sample. Thus when it determines repetitiveness it also determine uniqueness in the DNA. analysis is based on the reassociation kinetics of the completely denatured dsDNA. Cot value is equal to the product of initial concentration of the DNA subjected to reassociation (in Moles/li...
DNA Reassociation Kinetics - Part - 1

Repetitive DNA (Satellite DNA) The chromosome of prokaryotes almost exclusively contains DNA molecules with unique or non-repetitive DNA, i.e. each gene is present only once in the genome. But the chromosomes of eukaryotes are much more complex in this respect. Certain base sequences are repeated many more times in the haploid set of chromosome complement. DNA containing such repeated sequences...
